Seems like those all have their own group of enemies they're suitable for fighting.

EVA-01: For fighting Mobile Suits, EVA-units and other stuff that is roughly 18 meters tall.
Gunbuster: For fighting whatever you may find that is larger than EVA-01's size-category.
Tengen Toppa Gurren Lagann: For when you don't want to bother with looking for the enemy and just want to kill them. (not that hard, considering it fights on top of a galaxy and uses one to block two galaxies thrown at it like shurikens. AND it can survive having a Big Bang smashed against it)
