男の娘 is actually kind of a pun, since 娘 can also be pronounced "ko". It's also mimicking the structure of terms like 撲っ娘 or 眼鏡っ娘, bokukko and meganekko, to refer to a certain type of girl (in this case, one who is really a boy).
